BRA-1 is one of two C. elegans homologs of a human BMP receptor-associated molecule, BRAM1. BRA-1 interacts physically with the transforming growth factor-beta type I receptor DAF-1; the mutation bra-1(nk1) suppresses daf-1; and the bra-1 gene is coexpressed with daf-1 in amphid neurons such as ASK, ASI, and ASG. bra-1(nk1) also suppresses mutations in daf-7 and daf-14, and BRA-1 physically interacts with DAF-7 in vitro. bra-1 mutations have no effect on the phenotype of the null daf-1(e1287) mutation, or on that of the SMAD mutation daf-14(m77). BRA-1 has thus been proposed to exemplify a new class of proteins negatively regulating TGF-beta signalling by acting directly on the TGF-beta receptor. bra-1(nk1) homozygotes exhibit a 'head-lifting' phenotype.
