Functional description:
Protein LET-92; LEThal family member (let-92) [Source:RefSeq peptide;Acc:NP_502247] [Ensembl]; let-92 encodes a homolog of PP2AC, the catalytic subunit of protein phosphatase 2A (PP2A); let-92 was identified genetically in screens for essential genes on chromosome IV; LET-92 may regulate a number of signal transduction pathways as it is required for embryogenesis, larval development, Ras-mediated vulval development, and axonal guidance in the DD and VD neurons; let-92 interacts genetically with sur-6 which encodes a regulatory subunit of PP2A; LET-92 protein is bound both by SMG-2 and by POS-1, and also interacts in vitro with UNC-51 and UNC-14; LET-92 is expressed cytoplasmically in nearly all cells and strongly in many neurons; LET-92 is also seen on centrosomes and P granules; LET-92, and possibly other phosphatases, and the PAR-1 protein kinase regulate the MEX-5 gradient in the early embryo by controlling the association of MEX-5 with slow- and fast-diffusing RNA-containing cytoplasmic complexes. In two-hybrid experiments, LET-92 interacts with SMG-2 but not SMG-5 or SMG-7. In contrast, SMG-5 interacts with F48E8.5, a homolog of the the structural subunit of protein phosphatase 2A (PP2A). However, SMG-2 coimmunoprecipitates both with LET-92 and SMG-5, and thus may act as a bridge between LET-92 and F48E8.5 in vivo. Meanwhile, POS-1 binds LET-92 both in two-hybrid experiments and in an in vitro GST pull-down assay. let-92 alleles are generally larval lethal, while let-92(RNAi) has more severe phenotypes (Emb and Lvl), indicating a requirement of LET-92 for both embryonic and larval viability in mass RNAi assays.
